Development and assessment of an environmental DNA (eDNA) assay for a cryptic Siren (Amphibia: Sirenidae)

چکیده
Environmental DNA (eDNA) assays have become a major aspect of surveys for aquatic organisms in the past decade. These methods are highly sensitive, making them well-suited monitoring rare and cryptic species. Current efforts to study Rio Grande Siren southern Texas been hampered due nature these salamanders. Arid conditions further add difficulty studying this species, as many water bodies they inhabit ephemeral, sometimes constraining sampling short window after heavy rain. Additionally, sirens known cease activity reside underground when ponds begin dry or temperatures increase. Conventional require extensive trap-hours be effective, which is not always possible within required window. This presents development novel eDNA assay technique elusive species using conventional PCR Sanger sequencing compares results with simultaneous trapping at multiple sites assess relative effectiveness procedure. detection via was significantly higher all compared trapping, confirming utility detection. methodology gives promise future work assessing distribution status has potential use on other amphibians.

15 صفحه اولDevelopment and Validation of Environmental DNA (eDNA) Markers for Detection of Freshwater Turtles
Environmental DNA (eDNA) is a potentially powerful tool for detection and monitoring of rare species, including threatened native species and recently arrived invasive species. Here, we develop DNA primers for a suite of nine sympatric freshwater turtles, and use it to test whether turtle eDNA can be successfully detected in samples from aquaria and an outdoor pond. We also conduct a cost compa...
متن کاملDevelopment of a Sensitive Quantitative Competitive PCR Assay for Detection of Human Cytomegalovirus DNA
Accurate and rapid diagnosis of human cytomegalovirus (HCMV) disease in immunocompromised patients has remained as a challenge. Quantitative competitive PCR (QC-PCR) methods for detection of HCMV in these individuals have improved the positive and negative predictive values of PCR for diagnosis of HCMV disease. In this study we used QC-PCR assay, using a co-amplified DNA standard, to quantitate...
متن کاملDevelopment of Genetic Markers for Environmental DNA (eDNA) Monitoring of Sturgeon
EXECUTIVE SUMMARY: All living organisms shed DNA into the environment. Recent studies have demonstrated that environmental water samples can be effectively assayed for the DNA of target organisms. The DNA contained in these samples is commonly referred to as environmental DNA or eDNA. Because DNA assays can be sensitive to very small amounts of genetic material, eDNA surveys have the potential ...
